This innovative approach precisely targets PDAC, showcasing promising initial results in inhibiting tumor growth and extending survival in animal experiments. The research findings were published in the prestigious journal Nature Communications.
\r\n
\r\nAdvancing Precision Medicine: dtEVs and Targeted Therapies
\r\nLed by Professor and Yushan Scholar Li James Lee from NYCU's Institute of Biopharmaceutical Sciences, the team collaborated with Professor Chi-Ying F. Huang, a distinguished faculty member at NYCU's Institute of Biopharmaceutical Sciences, and Dr. Yan-Shen Shan, the Dean of the College of Medicine at NCKU, forming a cross-campus research team.
\r\n
\r\nThe study introduces a novel precision medical solution that utilizes a "nanopore electroporation technology platform" to transfect specific plasmid DNA into cells, releasing a substantial amount of engineered dtEV.
\r\n
\r\nThese dtEVs can carry high doses of therapeutic RNA nucleic acid drugs, with their extracellular vesicle surfaces adorned with artificially modified targeting proteins. These proteins can fuse with tissue-targeting peptides and bind with humanized monoclonal antibodies, achieving precise targeting of PDAC tumor cells and enabling deep penetration into cancer tissues.
\r\n
\r\n
\r\nThe research results indicate that dtEVs carrying therapeutic RNA nucleic acid drugs effectively suppressed PDAC tumors in mice. In experiments involving mouse-derived xenografts (CDX) or patient-derived xenografts (PDX) of pancreatic cancer, the combination of dtEVs with low doses of the chemotherapy drug gemcitabine successfully inhibited large tumors simulating late-stage pancreatic cancer, as well as intra-mouse metastatic lesions. This led to an extended survival period in the animal models.
\r\n
\r\nIndustrializing Breakthroughs: Towards Clinical Application
\r\nThe research team highlights the potential of this treatment method to produce nucleic acid biopharmaceuticals with specific targeting properties, delivering multiple therapeutic RNA drugs to large solid tumors. Currently, the team is actively working on industrializing this research breakthrough, with the goal of clinical application, and is seeking suitable biotech companies for technology transfer.
\r\n
\r\nPDAC, the most common subtype of pancreatic cancer, poses a significant challenge due to its poor treatment outcomes, primarily attributed to the high fibrosis and density of tumors. The presence of multiple gene mutations in pancreatic cancer further complicates the efficacy of single-drug treatments, leading to high recurrence rates. This innovative research offers a new ray of hope for more effective treatment options for pancreatic cancer patients.
\r\n
\r\n","liaisonper":null,"liaisontel":null,"liaisonfax":null,"liaisonemail":null,"docs":[],"images":[],"videos":[],"audios":[],"resources":[]},{"subject":"Position Opening Announcement","dataClassName":null,"pubUnitName":null,"posterDate":null,"updateDate":"2024-02-01","detailContent":"Position Opening Announcement
\r\n
\r\nProfessors/Associate Professors/Assistant Professors
\r\n
\r\nNational Yang Ming Chiao Tung University
\r\nInstitute of Biomedical Engineering
\r\nHsinchu, Taiwan
\r\n
\r\nInstitute of Biomedical Engineering invites applications for positions at all levels: Full Professor, Associate Professor, and Assistant Professor. Professor Tung-Hu Tsai from the Institute of Traditional Medicine (ITM) at National Yang Ming Chiao Tung University (NYCU) has confirmed through pharmacokinetics that Molnupiravir and its active metabolite, NHC, can penetrate the placental barrier to reach the fetus and achieve effective virus inhibition concentrations. This research was published in the scientific journal ‘eBioMedicine,’ which is part of the Lancet series.
\r\n
\r\nDuring the COVID-19 pandemic, there has been a lack of information on medication research. According to the interim guidelines for clinical management of novel coronavirus infections by the Ministry of Health and Welfare, prescribing physicians should only consider using Molnupiravir for pregnant women if the benefits outweigh the risks and must thoroughly inform pregnant women about the risks of using the medication during pregnancy. Even with this risk assessment, it remains entirely unknown whether the treatment reaches a therapeutic concentration for the fetus.
\r\n
\r\nThis research is part of the epidemic prevention scientific research center project led by Vice President Muh-Hwa Yang and was carried out in collaboration with the University of Cambridge in the UK.
\r\nProfessor Tung-Hu Tsai leads the research team, which used self-developed microdialysis probes to collect samples from the blood, placenta, amniotic fluid, and fetuses of pregnant mice. The concentrations of Molnupiravir and its active metabolites were analyzed using Liquid Chromatography with tandem mass spectrometry (LC-MS/MS). The research found that approximately 29% of the effective metabolite in the mother’s bloodstream reaches the fetus, 19% reaches the amniotic fluid, and 9% reaches the placenta. Furthermore, when combined with past in vitro research literature, it was discovered that the overall concentration of effective metabolites entering the fetus through placental tissue after administration is approximately only one-third of the concentration in the mother’s blood.
\r\n
\r\nProfessor Tung-Hu Tsai stated that the concentrations of the drug in the blood, placenta, fetus, and amniotic fluid of pregnant mice were all higher than the effective therapeutic concentrations, confirming it as an effective treatment regimen. The dosage administered achieves effective treatment concentration ranges in both the pregnant mother and the fetus. This scientific discovery holds important reference value for the use of medication by pregnant women infected with the COVID-19 virus and whether it can effectively treat infections in the unborn fetus.
\r\n
\r\n
\r\n
\r\n ","liaisonper":null,"liaisontel":null,"liaisonfax":null,"liaisonemail":null,"docs":[],"images":[],"videos":[],"audios":[],"resources":[]},{"subject":"【Academia Sinica】Allostery and substrate selectivity on ABC sterol transporters","dataClassName":null,"pubUnitName":null,"posterDate":null,"updateDate":"2023-09-13","detailContent":"Speaker Name: Dr. Jyh-Yeuan Lee ( University of Ottawa, Canada)
\r\nLectures: Institute of Biomedical Chemistry
\r\nSpeech: 2023-09-21 11:00-12:00
\r\nLocation: IBC R209
\r\nWebsite: https://www.sinica.edu.tw/en/Calendar_Content/57/3093","liaisonper":null,"liaisontel":null,"liaisonfax":null,"liaisonemail":null,"docs":[],"images":[],"videos":[],"audios":[],"resources":[]},{"subject":"【Academia Sinica】Use of Engineered Intrinsically Disordered Proteins in Biosensing Applications","dataClassName":null,"pubUnitName":null,"posterDate":null,"updateDate":"2023-08-22","detailContent":"Speaker Name: Dr. Gabriel P. A total of nine artist teams focuses on the emerging trends of interpreting tradition with new technology, digitalizing biomimetic simulation, and juxtaposing virtual and real space. By virtue of the interactive multimedia, participants can explore the virtual world intertwined with the physical world, and be able to embody the concept of ‘Heterosymbiosis’.
\r\nThe 3 sections
\r\nFuture Puppet Show uses technologies such as VR, robotic arms, eye movement tracking, and motion capture to reinterpret and enhance the abstract beauty of traditional culture through technology and digital translation.
